What you will  need:
1.)  MP3 player like Windows Media Player, winamp etc.
2.)  An MP3 to convert to a ringtone.

What to do:

1.)  Open the MP3 in your player and hit play, making sure you have adjusted your views so that the song's elapsed time is showing.
2.)  Navigate to the point on the MP3 you want the ringtone to begin, and remember that time or write it down.
3.)  You have up to 20 seconds to use as a ringtone, so listen and watch the elapsed time to find a good cut off point in the song.  Keep in mind the last second will automatically fade out.
4.)  Go to your Ringtone System Control Panel and fill out the form on that page, using the start time and length you wanted.
5.)  Hit upload and you are done!

Tips for best results:

Due to limits on the speakers of cellphones, try and choose parts of the song that aren't heavily distorted.

Try and keep the ringtone as short as possible since this allows for easier transfer to cellhpones.

If your MP3 is fairly large in size consider first trimming it down to not much longer than you want the ringtone to be, this will greatly speed up creation time.
